North Shore University Hospital Research Corporation (Manhasset, NY) has patented a method for using non-primate mammalian cell lines having substantially no endogenous retroviral sequences as producer and packaging lines for preparation of human serum-resistant retroviral vector particles with improved safety, for use in gene therapy applications.
The patent describes an AAV manufacturing process in which numerous growth conditions used to culture the production cells are controlled in a manner that leads to the release of AAV vector particles into the cell culture medium without the need for breaking open the host cell membrane.
In a specific embodiment, the vector particles are retronectinimmobilized or ultracentrifugation-concentrated retroviral vector particles pseudotyped with the feline endogenous retrovirus (RD114) envelope protein .
