Then, 25 [micro]L of acetylthiocholine iodide (Electrophorus electricus, Sigma Aldrich) 1000 U [mL.sup.-1]; the reaction mixture, 125 [micro]L of 5'.5-dithio-bis (2- nitrobenzoate) (DTNB, Sigma D8130) (3 mM) and 50 [micro]L of Tris/HCl (0.1% m/v, pH=8) containing bovine serum albumin was added to each well.
Briefly, in the performed assay 20 [micro]L of 0.22 U/mL AChE from Electrophorus electricus was added to the wells containing 20 [micro]L of tested compounds (100 and 75 [micro]M in methanol or DMSO), 100 [micro]L of 3 mM 5,5'-dithiobis(2-nitrobenzoic acid) (DTNB), and 20 [micro]L of 15 mM acetylthiocholine iodide.
Electric eel (Electrophorus electricus) AChE and horse serum BChE were used, while acetylthiocholine iodide (AChI) and butyrylthiocholine chloride (BChI) were employed as substrates of the reaction.
