Thiamine is a component of thiamine pyrophosphate, which participates in oxidative decarboxylation of pyruvic acid and [alpha]-ketoglutaric acid.
Thiamine, in the form of thiamine pyrophosphate (TPP), becomes an essential cofactor in glycolysis and mitochondrial oxidative decarboxylation of carbohydrates for energy formation [1, 3].
The enzyme PDH requires cofactor thiamine pyrophosphate. Thus, in case of thiamine deficiency, lactate cannot be metabolized to pyruvate and accumulates in large amounts resulting in severe lactic acidosis.
