Initially, 40 decamer primers from two kits (20 from kit A and 20 from kit B) of random sequence (Operon Technologies, Inc., Alameda, CA, USA) were screened on a sub sample of two randomly chosen fish to test their suitability for amplifying mud eel RAPDs that could be accurately scored.
The main objective of our study was to investigate intra-specific genetic variation in mud eel. Considering different parameters like proportion of polymorphic loci, intra-population Similarity Indices (SI), gene diversity (h) [15] and Shannon's Information Index (I), we found some degree of genetic diversity in the collected samples from a localized area that reflects the fact that the species has still not experienced genetic deterioration.
