To induce myotubule formation prior to performing glucose uptake assays, C2C12 cells were cultured for an additional two days in DMEM containing 2% horse serum (Highveld Biological, Johannesburg, South Africa) at 37[degrees]C in humidified air and 5% [CO sub.2].
The satellite cells readily respond to regenerative cues such as exercise, or injury by proliferating into myoblasts, which will divide a limited number of times before terminal differentiation and fusion to form multinucleated myotubules (Wager and Conboy, 2005).
Researchers found that inhibitors of the enzyme deacetylase can switch the pathway of myoblasts (i.e., muscle precursor cells) from simply reproducing themselves to becoming mature cells that form myotubules (i.e., muscle fibers).
