Blood-Brain Barrier (BBB) research embodies the true meaning of a "translational model" of neuroscience wherein breakthroughs in basic neuroscience are delivered to the clinic and require an agent delivery strategy and/or the ability to target specific areas of the brain. Long recognized as "the A in ADME", pION has commercialized PAMPA and developed applications that are highly correlated with Caco-2, gastrointestinal tract (GIT) and blood-brain barrier (BBB) permeation. Two accessory mechanisms are also suggested to be involved in MCS, one involving nitric oxide and the other peroxynitrite:nitric oxide inhibition of cytochrome P450 activity and peroxynitrite-mediated increased permeability of the blood-brain barrier (BBB) (Pall 2002b; Pall and Satterlee 2001). |
