Fure-2-loaded muscle strips were held horizontally in the organ chamber of a fluorimeter (CAF-110; Jasco, Tokyo, Japan) filled with KH solution.
At the end of each annealing step, the fluorescent signal of each reaction was measured at a wavelength of 530 nm with the LightCycler fluorimeter.
The measurements will be carried out using a conventional fluorimeter consisting of a monochromator with a CCD detector at the exit slit, and a source consisting of a cuvette with a illumination region parallel to the entrance slit of the monochromator.
