Fure-2-loaded muscle strips were held horizontally in the organ chamber of a fluorimeter (CAF-110; Jasco, Tokyo, Japan) filled with KH solution. At the end of each annealing step, the fluorescent signal of each reaction was measured at a wavelength of 530 nm with the LightCycler fluorimeter.
