[beta]-D-glucosidase activity was chosen as a representative C-acquiring enzyme because this enzyme is involved in hydrolysis of cellobiose, the main product in hydrolysis of cellulose by cellulases [20], and it was measured with p-nitrophenyl-[beta]-D-glucopyranoside as the substrate in a modified universal buffer (MUB) (tris(hydroxymethyl)
aminomethane, 2.42 g; maleic acid, 2.32 g; citric acid, 2.8 g; boric acid, 1.26 g; and distilled water, 1 L) at pH 6.0 [20].
Liver samples were homogenized in 0.1 mol/L tris (hydroxymethyl)
aminomethane buffer at 4[degrees]C, pH 7.4, to make a 10% (w/v) homogenate, using a polytron homogenizer for 5 min and a sonic homogenizer for 3 min.
The main ion concentrations of the SBF were nearly equal to that of human plasma and the pH was adjusted to 7.4 by trihydroxymethyl
aminomethane (Tris) and 1.0mol/LHCl solution [17].
Soybean lecithin (Calbiochem), K[H.sub.2]P[O.sub.4], [Na.sup.2]HP[O.sub.4], peptone, HCl, [H.sup.2][O.sup.2], luminol (5-amino-2, 3-dihydrophthalazine-1, 4-dione), and Tris (hydroxymethyl
aminomethane base) were purchased from Merck (Germany).
Rats lower brainstems were homogenized using lysis buffer containing 30 ml tris(hydroxymethyl)
aminomethane hydrochloride (pH 7.5), 250 mL sucrose, 150 mL sodium chloride, 1.0 mL dichlorodiphenyltrichloroethane, and a protease inhibitor cocktail (Sigma; St.
Tris (tris hydroxymethyl
aminomethane) buffer solution with an alkali buffering function was used in this study.
The mixture was left for 30 min for complete dissolution at room temperature and centrifuged at 12,000 rpm for 30 min; the supernatant fluid was decanted, and the precipitate was dissolved in 10 mm of TRIS (oxymethyl)
aminomethane buffer, pH 9.0.
