Swedish pharmaceutical company Calliditas Therapeutics AB (publ) revealed on Tuesday the receipt of US Food and Drug Administration (FDA) orphan drug designation (ODD) for the treatment of primary biliary cholangitis (PBC), previously known as primary biliary cirrhosis.
The following combinations of MAS with skin manifestations have been so far reported in literature5-7: [i] alopecia universalis, pemphigus vulgaris and insulin-dependent diabetes mellitus; [ii] pemphigus along with autoimmune thrombocytopenia, hemolytic anemia and hepatitis as a part of type II MAS6; [iii] psoriasis, primary biliary cirrhosis and SLE/scleroderma/Sjogren's syndrome overlap; [iv] vitiligo, bullous pemphigoid and rheumatoid arthritis8; [v] bullous pemphigoid, primary biliary cirrhosis and vitiligo; [vi] vitiligo, alopecia areata and ulcerative colitis.
Several associations between autoimmune hepatitis, and polymyositis/dermatomyositis with other diseases have been documented: with thyroiditis and antiphospholipid syndrome; systemic sclerosis and cerebral vasculitis; primary biliary cirrhosis and thrombocytopenic purpura; myasthenia gravis and thymoma; membranous nephropathy and Sjogren's syndrome; and, finally, with systemic sclerosis and sarcoidosis of mediastinal lymphoid nodules (3-8).
Initially cholestasis was thought to occur as a complication of cholestasis (primary biliary cirrhosis has an osteoporosis prevalence of 15-50%), but later abnormal bone mineral density was reported in other liver diseases as well including: viral cirrhosis (20-53%),primary sclerosing cholangitis (50%), or alcoholic cirrhosis (30%)11.

Conatus Pharmaceuticals announced the publication of results from preclinical studies of its pan-caspase inhibitor emricasan, demonstrating improvements in portal hypertension and survival in two bile duct ligation mouse models of induced secondary biliary cirrhosis. In the first of the two current mouse studies, designed to evaluate treatment effect, emricasan decreased fibrosis, reduced portal hypertension and improved survival in mice with cirrhosis induced by bile duct ligation.
